Assay principle
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Histone H1 (H1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Histone H1 (H1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Histone H1 (H1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Histone H1 (H1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Precision of the test
Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level Histone H1 (H1) were tested 20 times on one plate, respectively. ELISA's stability
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. Description
A microtiter plate (spelled Microtiter is a registered trade name in the United States) or microplate or micro well plate or multiwell, is a flat plate with multiple "wells" used as small test tubes. The microplate has become a standard tool in analytical research and clinical diagnostic testing laboratories. A very common usage is in the enzyme-linked immunosorbent assay (ELISA), the basis of most modern medical diagnostic testing in humans and animals. A microplate typically has 6, 24, 96, 384 or 1536 sample wells arranged in a 23 rectangular matrix. Some microplates have even been manufactured with 3456 or 9600 wells, and an "array tape" product has been developed that provides a continuous strip of microplates embossed on a flexible plastic tape.
